Like Cialis, Tadalis comprises of the active element Tadalafil which is the third member of the drug class phosphodiesterase type 5 (PDE-5) inhibitors. It is chemically and pharmacologically different from the previous 2 members Sildenafil citrate and Vardenafil. Tadalafil though, has similar mechanism of action like Sildenafil citrate and Vardenafil. It blocks the action of the enzyme PDE-5 which regulates the penile erections. Increased quantity of PDE-5 is known to cause restricted flow of blood in the penis which in turn leads to ED. Blockade of PDE-5 leads to increase in the levels of the biochemical nitric oxide and guanylyl cyclase which is an enzyme. Both this biochemical in synchronization, causes elevated synthesis of cGMP in the body near the penile region. The enzyme cGMP is known to cause vasodilation and is a potent vasodilator in the body. Hence, increased production of cGMP causes widening of blood vessels especially, the arteries that carry the blood to the penis from the heart. Also, under the influence of cGMP the smooth muscle below penis relax and allow the retention of more blood inside the penile arteries. This enhancement of blood flow to the penile region causes full-fledged penile erection which is sustained for a long time for satisfactory sexual intercourse.

Tadalis for use as needed

ED: Starting dose: 10 mg as needed prior to sexual activity. Increase to 20 mg or decrease to 5 mg based upon efficacy tolerability. Improves erectile function compared to placebo up to 36 hours post dose. Not to be taken more than once per day

Tadalis for once daily use

  • ED: 2.5 mg taken once daily, without regard to timing of sexual activity. May increase to 5 mg based upon efficacy and tolerability
  • ED and BPH: 5 mg, taken at approximately the same time every day

Usage instructions

  • This medicine may be taken without regard to food
  • To be swallowed as whole with water without crushing, chewing or splitting
  • A gap of at least 30 hours should be present between the two consequent doses of Tadalis

Use in specific populations

  • For patients with Mild or Moderate liver disease, doctor may recommend dosage adjustment
  • For patients with severe liver disease, use of Tadalis is not recommended.
  • In case of mild or moderate kidney damage, doctor may adjust the dose of Tadalis 20mg For patients with severe kidney failure and those on hemodialysis, not more than 5 mg every 72 hours should be taken.
  • Once daily use is not recommended in patients with severe kidney failure and those on hemodialysis.
  • DO NOT USE if sex is ill-advised due to heart diseases.
  • Use of Tadalis SX 20 mg with alpha-blockers, antihypertensives or considerable amounts of alcohol (five or more units) may lead to a severe decrease in blood pressure.
  • TADALIS intake is not recommended with alpha-blockers because of the risk of blood pressure lowering. Caution is advised when this medication is used as a treatment for ED in men on alpha-blockers.
  • Patients should stop this medicine and seek prompt medical attention in the event of sudden decrease or loss of hearing

Most common adverse reactions include headache, dyspepsia, back pain, myalgia, nasal congestion, flushing, and pain in limbs.

Get emergency medical help if allergic reaction to Tadalis is observed; symptoms of which includes:

  • Hives
  • Difficulty breathing
  • Swelling of face, lips, tongue, or throat
  • Stop and get medical help at once if you have nausea, chest pain, or dizziness during sex. You could be having a life-threatening side effect.
  • Stop using this medicine and call your doctor in case you observe:
  • A light-headed feeling
  • Painful erection that lasts longer than 4 hours (this can damage the penis)
  • Vision changes or sudden vision loss which could be a sign of non-arteritic anterior ischemic optic neuropathy (NAION)
  • Ringing in your ears or sudden hearing loss
  • Heart attack symptoms which include chest pain radiating to the left arm that is spreading to your jaw or shoulder, nausea, sweating.

Which drugs cannot be taken with Tadalis?

  • Drugs like nitrates, alpha-blockers, antihypertensives or alcohol can potentiate severe drop in blood pressure when given with this medicine.
  • Medicines that contain ketoconazole or ritonavir as active components can increase blood levels of Tadalis and dose adjustment might be required.
  • Drugs that contain rifampin as active ingredient decrease the blood concentration of Tadalis Tablets and hence, might degrade its action.

What should be done in case of Overdose?

  • Seek emergency medical attention or call the Poison control center if accidental or intentional overdose is suspected.
    • What if a dose of Tadalis is missed?

      • Since, this tablet has to be taken on as needed basis, there is less possibility that you might miss a dose.
      • If on a regular dosing schedule as prescribed by the doctor and if a dose of this medicine is missed, you should take it as soon as you recall.
      • If time is close to the nest dose skip the missed dose and wait for the time for the next dose.
      • Taking double dose in order to make up for missed dose is not at all recommended.
      • A gap of at least 30-hours should be kept between two doses in order to avoid major side effects.
